We have shown, that the MAPK module can produce amplification, switch-like response and can be used to adapt to the input. The switch-like response can be optimized, if the first and the second layer operate at low activation. To maximize the amplification the cascade can exploit large slopes when the operating-range of the second step is at intermediate activation.
